    ❣️I think this plot is at the top of the list for the most important plot of 2025. This notion of identifying and targeting with drugs sparse “drivers” of cancer is not consistent with actual pan-cancer gene expression multivariable analysis reality at all!

    Rarely have I seen an article miss the mark this badly 🤯 Misguided calls for more “real world” observational evidence should be replaced by calls for more efficient RCTs; patients and society deserve inference based on RCTs, nothing less

    Oncology drug exclusion principle: the more targeted the drug, the faster evolutionary escape makes failure a certainty; the less targeted are bounded by nonspecific cytotoxic chemo, which faces less escape, but cure is limited by exposure to rest of the body to high deg toxicity
